保护剂和再水化剂对冷冻干燥保存的生防菌YT11的 存活率及生防效果的影响

Impact of Protective Agents and Rehydration Media on Viability and Efficacy of the Freeze-dried Biocontrol Agent Pantoea ananatis Strain YT11

摘要: 本文研究了多种保护剂和再水化剂对生防菌成团泛菌Pantoea ananatis菌株YT11在冷冻干燥保存中存活率的影响,比较了不同保存温度对YT11贮藏稳定性和生防活力的差异。结果表明,5%蔗糖是最佳的保护剂,冷冻干燥后菌株YT11存活率达90%以上。以5%蔗糖作保护剂时,5%脱脂奶粉为最佳再水化剂,菌株YT11复苏率达100%。贮藏试验结果表明,4℃贮存的冷冻干燥粉剂的菌株存活率显著高于22℃下的存活率。对保存24周后的菌剂进行温室生防试验,结果显示,4℃和22℃下保存的菌剂对番茄青枯病的防效分别达到71.57%和50.98%。因此,以5%蔗糖为保护剂和5%脱脂奶粉为再水化剂,对成团泛菌菌株YT11进行冷冻干燥处理,再将冷冻干燥后的菌株于4℃贮存,可有效保证生防菌的生防活力。

Abstract: The effect of different protective agents and rehydration media on the viabilities of the biocontrol agent Pantoea ananatis strain YT11 subjected to freeze-drying, its subsequent survival rates during storage under different temperatures, and the biocontrol efficacies of the freeze-dried cells were investigated. Results indicated that 5% sucrose gave the greatest protection to YT11 with cell viability more than 90% during freeze-drying, and 5% non-fat skim milk as rehydration media gave the highest survival rate reaching 100%. Freeze-dried cells stored at 4℃ survived more than those at 22℃. YT11 powder formulation stored for 24 weeks at 4℃ and 22℃ gave control efficacies by 71.57% and 50.98%, respectively.
