苏云金素发酵培养基的优化设计

Optimized Fermentation Medium for the Production of Thuringiensin

摘要: 采用Plackett-Burman 设计对出发培养基各种成分影响苏云金芽孢杆菌中华亚种CT43 发
酵合成苏云金素的效应进行评价,筛选出有显著影响效应的因素为葡萄糖和黄豆饼粉。利用
最陡爬坡路径试验逼近最大苏云金素产出区域,找到后续试验中心点。综合中心组合设计和
响应面分析法确定显著因素的最佳浓度。优化后培养基组成为(g/ L) :葡萄糖22.8g、黄豆饼粉32.5g、酵母抽提物3.0g、蛋白胨4.5g、玉米浆3.0g、KH2PO4 2.0g、MgSO4·7H2O 01.5g、MnSO4·H2O 0.02g ,苏云金素产量可达3.2g/ L ,比出发培养基提高2 倍以上。

Abstract: Experiments was designed to optimize the fermentation medium of thuringiensin produced by Bacillus thuringiensis subsp. chinensis CT43. Initially , Plackett-Burman design was applied to evaluate the effectiveness of the related influential factors ; the selected ones that affected the fermentation were glucose and soybean powder. Then the steepest ascent experiment was adopted to approach the optimal re-gion of the medium composition. At last , the optimum composition of the fermentation medium was ob-tained by central composite design and response surface methodology , that was : 22.8 g·L - 1 of glucose ,32.5 g·L - 1 of soybean powder , 3.0 g·L - 1 of yeast extract , 4.5 g·L - 1 of peptone , 3.0 g·L - 1 of corn mash , 2.0 g·L - 1 of KH2PO4 , 0.15 g·L - 1.of MgSO4·7H2O and 0.02 g·L - 1 of MnSO4·H2O. The final yield of thuringiensin was 3.2 g·L - 1 .After optimization , the yield of thuringiensin production was in creased over twice compared with that in the original medium.
