环境因素驱动纹彩猎蝽雌虫肠道菌群结构与多样性显著差异

Structure and Diversity of Gut Microbiota in Female Euagoras plagiatus Depend Largely on Environmental Factors

摘要: 肠道细菌群落在宿主昆虫的整个发育阶段中发挥着关键作用。本研究基于PacBio平台,利用16S rDNA全长扩增子测序技术,系统解析了不同生存环境下纹彩猎蝽Euagoras plagiatus雌虫肠道细菌的群落特征。结果表明,野生型(Y)与实验室饲养型(Z)纹彩猎蝽雌虫肠道中分别检测到650个和239个操作分类单元(OTU),共有OTU为218个。Alpha和Beta多样性分析均表明,野生型个体的肠道细菌群落具有更高的丰富度与多样性。此外,通过细菌分离技术,成功从纹彩猎蝽雌虫肠道中分离出19株细菌,分属6个不同种。荧光定量PCR (RT-qPCR)分析进一步揭示,内生细菌在野生型个体中的丰度与饲养型存在显著差异。本研究揭示了环境因素对纹彩猎蝽雌成虫肠道细菌群落组成及多样性的影响,为其环境适应机制的深入理解提供了重要理论依据。

Abstract: The gut bacterial community plays a crucial role throughout the developmental stages of host insects. In this study, based on the PacBio platform and using full-length 16S rDNA amplicon sequencing technology, we systematically analyzed the gut bacterial community characteristics of female Euagoras plagiatus under different living environments. The results revealed that 650 and 239 operational taxonomic units(OTUs) were detected in the guts of wild-type(Y) and laboratory-reared(Z) female E. plagiatus, respectively, with 218 OTUs shared between the two groups. Both Alpha and Beta diversity analyses indicated that the gut bacterial community of wild-type individuals exhibited higher richness and diversity. Furthermore, through bacterial isolation techniques, 19 bacterial strains belonging to 6 different species were successfully isolated from the gut of female E. plagiatus. Quantitative real-time PCR(RT-qPCR) analysis further revealed significant differences in the abundance of endogenous bacteria between wild-type and reared insects. This study elucidates the influence of environmental factors on the composition and diversity of the gut bacterial community in female E. plagiatus adults, providing an important theoretical basis for a deeper understanding of their environmental adaptation mechanisms.
