稻纵卷叶螟颗粒体病毒亚致死作用对宿主生长、繁殖和生理生化的影响

  

Sub-lethal Effect of Cnaphalocrocis medinalis Granulovirus to the Growth, Reproduction and Physiology of Host

摘要:

稻纵卷叶螟颗粒体病毒(CnmeGV)是感染稻纵卷叶螟的专性杆状病毒。明确CnmeGV亚致死作用对稻纵卷叶螟生长发育、繁殖能力和生理生化的影响,为应用CnmeGV防治稻纵卷叶螟提供依据。CnmeGV 1×105 OB/g浓度处理3龄稻纵卷叶螟幼虫10 d后存活幼虫虫体内仍普遍检测到CnmeGV的存在,从而产生亚致死作用。亚致死CnmeGV处理对稻纵卷叶螟的生长发育产生明显影响,主要表现在变态发育的化蛹、羽化和产卵过程,存活个体蛹重减轻了15.95%,羽化率下降了24.81%,产卵量减少41.5%,差异显著。同时雌雄性比从1.11下降到0.82。CnmeGV亚致死处理诱导幼虫能量代谢增强,中肠消化酶中淀粉酶、脂肪酶活性显著提高,而解毒酶中羧酸酯酶活性显著下降,谷胱甘肽转移酶活性显著提高。CnmeGV亚致死作用影响到虫体激素分泌,幼虫体内蜕皮蜕皮激素、保幼激素水平较对照分别下降了4.59和5.62倍。CnmeGV亚致死处理雌成虫卵黄蛋白含量下降了40.63%,可能是导致产卵量降低的主要因素。

Abstract:

 Cnaphalocrocis medinalis granulovirus, CnmeGV, is a specific pathogenic baculovirus of rice pest Cnaphalocrocis medinalis. The objective of this study is to analyze the sub-lethal effects of CnemGV to the growth, reproduction and physiology of C. medinalis, to provide a theoretical basis for the application of baculovirus for pest control. CnmeGV was still detected in the survival larvae 10 days later after 3rd instar larvae of C. medinalis treated with sub-lethal dose of CnmeGV at high frequency, resulting in sub-lethal effect to C. medinalis. Sub-lethal treatment of CnmeGV did great impaction on the development of C. medinalis, especially in the metamorphosis process of pupation, eclosion, and oviposition. The pupa weight decreased by 15.95%, eclosion rate reduced by 24.81%, oviposition dropped by 41.5%, significantly different from that of control. The ratio of female to male also declined from 1.11 to 0.82. Sub-lethal effect of CnmeGV enhanced the energy metabolism of larvae, with the actives of amylase and lipase raised remarkably. Among detoxifying enzymes, the active of GST was improved, while ACT level dropped significantly compared to that of control. Sub-lethal of CnmeGV influenced the secretion of hormones. The levels of juvenile and molting hormone declined by 5.62 and 4.59 folds respectively. Vitellogenin content of females was detected to reduce about 40.63% in the sub-lethal treatment. This might affect the oviposition rate of adults.
