用血清学方法测定苏云金芽孢杆菌晶体蛋白含量

SEROLOGICAL ASSAYS FOR QUANTITATION OF THE SOLUBLE CRYSTAL PROTEIN OF BACILLUS THURINGIENSIS

摘要: 用苏云金芽孢杆菌HD-1、7216和CT-43菌株晶体蛋白的碱溶和胰蛋白酶酶解产物作抗原,制备相应的抗血清。用环状沉淀反应、双相单扩散反应和火箭免疫电泳等技术测定了苏云金杆菌几个菌株和制剂的晶体蛋白的含量。用环状沉淀反应测定晶体蛋白的最低量为0.22μg/mL,火箭免疫电泳为15μg/mL,双相单扩散反应为62μg/mL。生物测定的结果与几种血清学方法测定的结果有一定的相关性,菌株HD-1、7216和CT-43的伴胞晶体经碱溶和胰蛋白酶处理后对小菜蛾幼虫的LC50值分别为0.148、0.247和0.097μg/mL;经碱溶而未经胰蛋白酶处理的晶体蛋白对小菜蛾幼虫的LC50值分别为1.867、3.147和1.022μg/mL,两种处理方法对晶体蛋白毒力影响很大。环状沉淀反应测定苏云金芽孢杆菌晶体蛋白含量灵敏度比火箭免疫电泳高,操作简便而快速;双相单扩散法灵敏度与火箭免疫电泳接近,但操作更简便。

Abstract: The antigen and antiserum of crystal protein of Bacillus thuringiensis strains
HD-1, 7216 and CT-43 which were solubilized in alkali or trypsin were
prepared. The circular precipitation reaction, double direction single diffusion reacton, and rocket immunoelectrophoresis were used to quantitate those soluble crystal preteins. The minimal detectable value was 0.25Lg/ml by the circular precipitation, the minimal detectable value was 62Lg/ml by double direction-single diffusion, and the minimal detectable value was 15Lg/ml by rocket immunoelect rophoresis.The result of bio-assay was relevant with that of serological methods. However, methods of serological methods were simpler, faster than bioassay. The circular precipitation was the most simple, the most rapid within the several serological methods. It was first success that the circular precipitation and the double direction-single diffusion were used to quantitate soluble crystal protein of B. thuringiensis.
