诱导子对黄曲霉生物合成乌头碱的诱导作用研究

Induction of Aconitine Biosynthesis in Aspergillus flavus by Inducers

摘要: 为探索不同诱导子对生物合成乌头碱产量的影响,本研究在黄曲霉Aspergillus flavus的液体发酵过程中分别加入金属诱导子( Co2+、Ag+和Cu2+)和化学诱导子(茉莉酸甲酯、水杨酸和硝普钠),以及酵母提取物、种子提取物和共生细菌发酵液等生物诱导子进行诱导试验。试验结果表明,Ag+、Cu2+和Co2+都对乌头碱的生物合成有促进作用,其中Ag+浓度为50 μmol/L时,乌头碱产量与对照组相比提高了10倍。化学诱导子MJ和SA诱导效果显著,其中100 μmol/L MJ的加入能显著提高乌头碱的产量,产量为20.74 mg/L,为对照组的11倍。而酵母提取物、种子提取物和共生细菌发酵液对乌头碱的产量诱导效果均不显著。本论文探究了不同诱导子对黄曲霉生物合成乌头碱的诱导作用,进而明确不同诱导子及不同诱导浓度对乌头碱生物合成产量的影响。

Abstract: In order to explore the effect of different inducers on the yield of biosynthesis aconitine,the present study was carried out in the liquid fermentation of Aspergillus flavus by adding metal inducers (Co2+,Ag+ and Cu2+) and chemical inducers (Methyl Jasmonate,MJ,Salicylic Acid,SA and Sodium nitroprusside,SND),as well as YE (Yeast extract),SE (Seed extract) and Bac (Symbiotic bacterial fermentation broth).The results showed that Ag+,Cu2+ and Co2+ all could promote the biosynthesis of aconitine,in which Ag+ at 50 μmol/L increased the aconitine yield by 10-fold compared with the control.Induction effects of the chemical inducers MJ and SA were significant,in which the addition of 100 μmol/L MJ significantly increased the production of aconitine with a yield of 20.74 mg/L,which was 11-fold higher than that of the control,while YE,SE and Bac did not have any significant effect on the biosynthesis of aconitine by Aspergillus flavus.
