球孢白僵菌在玉米苗期的定殖及其对玉米生理生化特性的影响

Colonization of Beauveria bassiana in Maize Seedlings and Its Effect on Their Physiological-biochemical Characteristics

摘要: 为明确球孢白僵菌D1-5在玉米苗期的定殖规律及其对玉米生长状态和生理生化特性的影响,采用盆栽控制试验,研究了两种浸种时间(12和24 h)对菌株D1-5在玉米苗期不同部位(根、茎、叶)的定殖偏好性的影响,同时研究了几种生长指标对不同浸种时间的响应,分析了抗性相关防御酶活性(出苗14 d)变化情况。结果表明,叶片中菌株D1-5定殖检出率最高,达到40%,茎部其次,定殖检出率为10%,玉米根系未检测出菌株D1-5;各检测时期菌株D1-5处理组均高于对照组,差异显著,其中玉米出苗19 d生长势增长率最高,达到5.34%,出苗21 d叶面积增长率最高,达到13.71%,出苗28 d地上生物量增长率达到27.2%;菌株D1-5处理后叶片中抗性相关防御酶——苯丙氨酸解氨酶(PAL)、多酚氧化酶(PPO)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)活性均呈升高趋势,SOD活性增加最高,达到24.90%。菌株D1-5浸种对玉米苗期的生长发育有一定的促进作用,其中浸种12 h更有利于玉米幼苗生长,玉米相关防御酶的活性变化同球孢白僵菌-玉米之间的互作存在一定的相关性。

Abstract: Colonization of Beauveria bassiana in maize seedlings and its effect on their physiological and biochemical characteristics was investigated by examining colonization positions (leaf, stalk and root) under 2 seed soaking durations (12 and 24 h), and measuring plant growth characteristics and activities of some resistance defense enzymes in 14 d old seedlings. Endophytic colonization of B. bassiana in leaves (40%) was greater than in stalks (10%), and no endophytic colonization in roots was found. Significant effects of seed soaking durations on bio-physiological parameters of maize seedlings were detected. Plant height was increased by 5.34% in 19 d old seedlings, and leaf area and aboveground biomass by 13.71% and 27.2% in 21 d and 28 d old seedlings, respectively. Activities of some resistance defense enzymes, including phenylalnine ammonialyase (PAL), polyphenoloxidase (PPO), superoxide dismutase (SOD), catalase (CAT) and peroxidase (POD), were enhanced, with increase by 24.90% for SOD. Our study suggests that B. bassiana may have positive effects on plant growth, especially for seed soaking treatment for 12 h. In addition, changes in activities of defense enzymes may be dependent on the interactions between B. bassiana and the seedlings.
