温度对2株伯氏致病杆菌生长速度和致病力的影响

Effects of Temperature on Growth and Pathogenicity of Two Strains of Xenorhabdus bovenii

摘要: 为了明确温度对昆虫病原线虫共生细菌-伯氏致病杆菌Xenorhabdus bovenii的生长及致病力的影响及其最佳培养条件,该试验通过平板和发酵液培养法,在不同温度处理条件下,对已分离鉴定的2株伯氏致病杆菌 0641TY和0657L品系进行培养,研究其生长速率和其对大蜡螟Galleria mellonella的血腔毒力及胃毒毒力。结果表明:2株伯氏致病杆菌在平板中培养时,30 ℃条件下伯氏致病杆菌0641TY和0657L的菌落平均直径分别为1653.34和834.84 μm,显著大于其他温度;发酵液中培养时,30 ℃条件下2株菌分别于6和12 h时达到对数生长期,生长速率均显著高于其他温度;30 ℃培养的2株伯氏致病杆菌发酵液对大蜡螟幼虫有较高的血腔毒力,在侵染72 h校正死亡率均达到100%,而胃毒毒力较弱,侵染96 h致死率分别为50%和46.6%。因此,30 ℃有利于2株伯氏致病杆菌的生长和保持其毒力。

Abstract: To determine the influence of temperature on the growth and virulence of the symbiotic bacteria, as well as the optimal culture conditions, the experiment was conducted by plate and fermentation broth culture method under different temperature treatment conditions. Two strains of X. bovenii 0641TY and 0657L were cultured to study their growth rate and their hematotoxicity and gastrotoxicity to G. mellonella. The results showed that the average colony diameters of the X. bovenii 0641TY and 0657L with plate culture were 1653.34 and 834.84 μm respectively at 30 ℃. The two strains reached logarithmic growth stage at 6 and 12 h respectively at 30 ℃, and the growth rate was significantly higher than that at other temperatures.. The two strains cultured at 30 ℃ showed high gastrotoxicity to the larvae, and the corrected mortality rate reached 100% after 72 h of infection, while the hematotoxicity was weak, the mortality rate was 50% and 46.6% respectively after 96 h of infection.Therefore, 30 ℃ was beneficial to the growth and virulence of the two strains of X. boveni.
