产蛇孢菌素A禾长蠕孢REMI突变体的制备及其筛选

Screening of Helminthosporium gramineum Ophiobolin A Producing Mutants Created with Restriction Enzyme Mediated Integration

摘要: 蛇孢菌素A具有除草、抗菌等广泛的生物活性,在农业领域具有重要的应用价值。本研究采用限制酶介导整合法(REMI)将外源质粒pSH75转化产蛇孢菌素A真菌禾长蠕孢菌获得一系列转化子,进一步采用发酵液抑菌法和稗苗生测法筛选弱毒力菌株,最后用HPLC方法探测弱毒力菌株中蛇孢菌素A产量。试验共获得6株弱毒力菌株,其中B014和B016为蛇孢菌素A合成缺失菌株,H002和B018为蛇孢菌素A毒素合成降低的突变株。为下一步蛇孢菌素A毒素合成相关基因的克隆和蛇孢菌素A基因调控奠定基础。

Abstract: Ophiobolin A is sesterterpenoid-type phytotoxin with potential for weeds and diseases control. The restriction enzyme-mediated integration (REMI) was used to introduce the plasmid pSH75 into the ophiobolin A-producing Helminthosporium gramineum. Mutants with lower toxicity were selected with bioassay of the cell-free cultural filtrates. And the amount of ophiobolin A of lower toxicity mutants was detected with HPLC. In comparison with wild-type strain, the cell-free cultural filtrates of 6 REMI transformants showed significantly less inhibition on mycelial growth of plant pathogen Rhizoctoni solani, and therefore were categorized into lower toxicity mutants. Among the 6 strains, strains B014 and B016 produced no detectable ophiobolin A as measured with HPLC. Strains H002 and B018 produced less ophiobolin A in comparison with wild-type strain. These mutants might be useful for tagging and cloning genes related to ophiobolin A production in the future. This research also suggested REMI as a potential approach for improving the production of ophiobolin A by H.gramineum via genetic engineering.
