蝉棒束孢交配型基因的分析和检测

摘要: 蝉棒束孢Isaria cicadae为药用真菌蝉花的无性型,在防治同翅目害虫上具有重要作用。在自然状态下蝉棒束孢多以孢梗束状态存在,其子实体阶段极为少见。交配系统是真菌有性生殖过程中的决定因素,因此可从交配型的角度探究蝉棒束孢有性子实体稀少的原因。本研究运用RACE技术首次从蝉棒束孢中克隆出MAT1-1-1MAT1-2-1全长cDNA序列,并进行生物信息学分析。根据cDNA序列设计并优化得到蝉棒束孢交配型鉴定引物,并对采集自安徽地区40株蝉棒束孢交配型进行鉴定,结果表明交配型MAT1-1菌株14株、MAT1-2菌株26株,并没有检测到同时含有两种交配型或两种交配型全部缺失的现象,因此蝉棒束孢有性型子实体几无发生的现象并不完全归因于交配型因素。

Abstract: The filamentous fungus Isaria cicadae is known as the anamorph of medicinal fungi Cordyceps cicadae and is widely used for biological control against homoptera pests. Although I. cicadae is widespread in the wild, Isaria cicadae is present mainly in the form of coremium and finding sexual development of this fungus is extremely difficult because of its obscure sexual stages. The mating system of fungi is a determining factor for sexual reproduction, but mating-type (MAT) genes of I. cicadae have not been previously investigated. Therefore, the presence and functionality of MAT genes in the fungi were investigated to assess the potential for sexual reproduction in this study. Firstly, the full-length cDNA of MAT1-1-1 and MAT1-2-1 were cloned from I. cicadae using RACE technology, and the characteristics of the genes were analyzed. Furthermore, based on the full-length cDNA, PCR primers set for the MAT1-1-1 and MAT1-2-1 were designed and optimized, respectively. And the mating type of 40 strains from Anhui province were determined to be MAT1-1 (14 strains) were identified while the others were MAT1-2 (26 strains), with no deletion or a dominant mating type strains in the distribution. These results suggested that other elements, such as growing environment or hereditary factors, may contribute to the obscure sexual fruiting-body instead of mating type.
