贝莱斯芽胞杆菌ZJ1油悬剂的研制及其田间防病促生效果

Preparation of Biocontrol Agent for Bacillus velezensis ZJ1 Oil Suspension and their Plant Growth-promoting and Biocontrol Properties in Field

摘要: 采用平板对峙法筛选出1株对番茄早疫病菌Alternaria solani和番茄灰霉病菌Botrytis cinerea均具有良好抑菌活性的贝莱斯芽胞杆菌Bacillus velezensis ZJ1。采用冷冻干燥法及生物相容性试验进行助剂筛选获得菌剂配方,并进行ZJ1油悬剂的田间防病效果及促生作用研究。结果表明,菌株ZJ1油悬剂的最佳配方为10%菌粉(6.0×1010cfu/g)、7%(v/v)吐温80、50 μg/mL两性霉素、2% CaCl2、1% SDS、1% CaCO3、0.5%木质素磺酸钙、5%糊精,余量用花生油补足,芽胞含量为5.75×109 cfu/mL。油悬剂各项指标均符合国家标准。贝莱斯芽胞杆菌ZJ1油悬剂200倍稀释液对A. solani引起的早疫病和B. cinerea引起的灰霉病防治效果达90%以上,具有良好的保护效果和治疗效果,有明显的促生作用,可使番茄植株茎粗显著增大。本研究制备的防病促生菌剂环保高效,效果稳定,为其生产及田间应用奠定基础。

Abstract: A strain of Bacillus velezensis ZJ1 with good inhibitory efficiency against tomato early blight and gray mold caused by Alternaria solani and Botrytis cinerea was accquired by using the dual culture. The freeze drying method and biocompatibility test were used to select the optimal formula of bacterial agent, the field efficacy and the growth-promoting effect of ZJ1 oil suspension was determined. The optimum formulation of ZJ1 strain oil suspension was 10% bacteria powder (6.0×1010cfu/g), 7% (v/v) Tween80, 50 μg/mL amphotericin, 2% CaCl2, 1% SDS, 1% CaCO3, 0.5% calcium lentisulfonate, 5% dextrin, peanut oil to complement to 100%, the spores content is 5.75×109cfu/mL. All indexes were in line with national standards. The B.velezensis ZJ1 oil suspension with the dilution of 200× had the best biocontrol effect on tomato early blight and gray mold caused by A.solani and B.cinerea, the protective and therapeutic effects can reach more than 90%, and has obvious growth-promoting effect, and could significantly increase the stem diameter. The preparation of this biocontrol agent is environmentally friendly and highly-efficient, and has stable effect, which lays a foundation for its production and application in fields.
