Transcriptional Regulation and Overexpression of glnA Gene in Bacillus thuringiensis

Abstract: Nitrogen metabolism is essential for bacterial survival, while glutamine and glutamate play an important role in bacterial nitrogen metabolism. The transcriptional regulation and overexpression of the glutamine synthetase gene of Bacillus thuringienesis was examined in this study. By constructing the glnR deletion strain and glnA promoter fusion with lacZ gene, the activity of glnA promoter was investigated in the mutant strain and wild type stain. The results showed that glnA gene was negatively regulated by transcriptional factor GlnR in the case of sufficient extracellular nutrition source. Overexpression of glnA gene in glnR mutant increased insecticidal activity compared to the wild type strain.
