Differential Expression of Aphis glycines Gene Aghsp75 in Response to Imidacloprid and Temperature Stress

Abstract: Heat shock proteins (HSP) play important roles in insect resistance to temperature and insecticide stress. A heat shock protein cDNA sequence of Aphis glycines was obtained by high throughput sequencing, which was a 2982 bp full-length sequence and contained a 2052 bp open reading frame. It encoded a polypeptide consisting of 683 amino acids. The isoelectric point of the polypeptide was about 6.30 and the molecular weight was about 77.8 kDa. The deduced amino acid sequence shared 99.12% identity with Aphis gossypii HSP75. The cDNA sequence was named Aghsp75. The GenBank accession number was MN068810. The relative expression of Aghsp75 was significantly increased after 3, 6 and 24 h induction by imidacloprid LC50 and 12 h induction by imidacloprid LC30, or after 6 h treatment at 6 ℃ and 3 h and 6 h treatment at 36 ℃. This study suggests that Aghsp75 may be involved in the stress resistance of A. glycines and provides theoretical support for the control of A. glycines by molecular biotechnology.
