3株苏云金杆菌噬菌体的核酸性质

摘要: 对形态结构基本一致的3株苏云金杆菌噬菌体的核酸性质及其限制性内切酶的酶切特性进行了比较研究。甲酰胺-细胞色素C法进行分子展层,发现其核酸均呈线状;核酸的热变性试验和S1酶的电泳结果表明,所有核酸均为双链;Hind 和BamH 酶对3株噬菌体核酸均不敏感;而它们均能被EcoR 切割成8个片段,被Hpa 切割成7个片段。经电泳法测定,其分子量均为48.0 8kb。

Abstract: The properties of nucleic acids of three Bacillus thuringiensis phages with similar morphology from CGVCC and their digested characteristics by restriction endonuclease were compared Nucleic acid of all of 3 phages were linear by molecular spreading after treatment of formamide cytochrome C, and were double strand by thermal denaturetion and electrophoresis of enzyme Sl DNAs of all 3 phages were not sensitive to restriction endonuclease HindⅢ and BamHⅠ,but were digested by EcoRⅠ and HpaⅡ into 8 and 7 fragments respectively The molecular weights of the phages' DNAs were about 48.08kb.
