小麦纹枯病拮抗细菌的筛选

SCREENING AND UTILIZATION OF ANTAGONISTIC BACTERIA FOR CONTROLLING SHARP EYESPOT OF WHEAT

摘要: 从小麦纹枯病、健叶鞘上分离到240个细菌分离物,通过细菌-纹枯菌对峙培养和含菌平板培养筛选出拮抗力较强的Rb2、Rb26等11个芽孢杆菌菌株。在对峙培养中,拮抗细菌的抑菌带达11.5~16.2mm,在含细菌培养基中,拮抗细菌产生明显的抑菌透明圈,宽度7.0~13.0mm,小麦纹枯菌不能生长甚至死亡。筛选到的Rb2和Rb26的代谢产物对小麦纹枯菌生长和菌核形成均有明显的抑制作用,抑制中浓度分别为2.8%和1.8%,且高温处理对其抑制活性无显著改变,抑制中浓度仍分别达3.0%和2.1%,但对菌核形成的抑制活性有所减弱。Rb2和Rb26菌液浸种对苗期纹枯病的防效分别达到71.8%和78.1%,代谢产物浸种的防病效果也达71.1%和72.8%。研究认为,Rb2和Rb26对小麦纹枯菌具有良好的拮抗作用,对苗期小麦纹枯病有明显的控制作用。

Abstract: 240 isolates were obtained from wheat sheath infected with Rhizoctonia
cerealis
using dilute plate technique and 11 of these isolates, Bacillus spp.
showed an tagonism to R. cerealis which causes wheat sharp eyespot. The inhibition zone of mycelial growth on PDA based on the fungus-bacteria pairing culture ranged from 6.0- 13.0 mm. The mycelial growth on the medium mingled
with bacterium was inhibited by 100% and having block zones in the plates of
Rb2, Rb13, Rb18, Rb23, Rb26 and Rb29. Rb2 and Rb26 were selected to be
goodan tagonists. The metabolites of Rb2 and Rb26 inhibited the mycelial
growth, their EC50 were 2. 8% and 1. 8% respectively. The boiled metabolites
also demonstrated the good inhibition, their EC50 were 310% and 2. 1% respectively. Sclerotia form ation as inhibited significantly by boiled metabolites, but less than the unboiled metabolites. The wheat seeds were soaked in the bacterial suspension and its metabolite, which was prepared from liquid culture and contained abou t 6. 8 × 108 colony forming units ( cfu )/m l for 24 hat 20℃, and seedlings were infected with inoculum of R. cerealis. The results demonstrated that Rb2 and Rb26 were effective for controlling wheat sharp eyespot and so were their metabolites, which implied that Rb2 and Rb26 may inhibited the disease with seed bacterization or by the metabolites.
