长枝木霉菌T115D诱导大豆叶片防御酶活性及疫病盆栽防治效果

Induction of Defense Enzymes Activities in Soyben and Control Effect of Phytophthora Root Rot in Flowerpot by Trichoderma longibrachiaum T115D

摘要: 采用分光光度和盆栽方法,分别测定长枝木霉菌T115D对大豆叶片防御酶活性的影响及大豆疫病的防治效果。结果表明,菌株T115D发酵液的不同处理均可诱导大豆叶片PAL、POD、PPO、SOD和CAT的活性增强,并且比单独接种疫霉菌处理的酶活持续时间长、峰值高;提前接种木霉有孢子发酵液后,第2 d再挑战接种大豆疫霉菌的处理诱导效果最好,PAL、POD、PPO和CAT 4种酶的活性在处理后第5 d达到最大值,分别是Petri培养液对照处理的2.5、6.6、4.8和3.5倍。菌株T115D发酵液不同处理均可降低大豆疫病的发生。发酵液喷雾处理中,提前1 d接有孢子发酵液,第2 d接大豆疫霉菌处理的防效最好,其防效为73.3%,显著高于其他处理;发酵液灌根处理中发酵液和疫霉菌同时灌根处理的防效最好,其防效为51.8%,显著高于其他处理。菌株T115D发酵液不同接种量对大豆疫病的防治效果,以接有孢子的发酵液15 mL最高,防效为达70.9%。

Abstract: The induction of defense enzymes activities and control effect of Phytophthora root rot by Trichoderma longibrachiaum T115D were measured by UV spectrophotometry and investigated by flowerpot experiments. The results showed that the activity of PAL, POD, PPO, SOD and CAT in the soyben leaves was improved after induction by fermentation broth of T115D. The five enzymes activities were of longer duration and higher peaks than those of the control treatment by Phytophthora sojae. When soyben leaves were first treated with spores in fermentation broth of T115D and then inoculated with P. sojae at the 2nd d, induced effect of defense enzymes activities was the best at the 5th day, and the activities of PAL, PPO, POD andCAT reached the maximum, which were 2.5, 6.6, 4.8 and 3.5 times as those of the petri culture liquid control, respectively. Different treatments of T115D fermentation broth could reduce the occurrence of phytophthora root rot. Among the spray treatments, when soyben leaves were first treated with fermentation broth of T115D by spray and then inoculated with P. sojaeat the 2nd d, control effect on phytophthora root rot was the best with the efficacy of 73.3%, which was significantly higher than that of other treatments. Among the root-dreaching treatments, when soyben were treated at the same time with spores in fermentation broth of T115D and P. sojae by root drenching, control effect was the best with the efficacy of 51.8%, which was significantly higher than that of other treatments. Among the treatments with different inoculation amount of T115D fermentation broth by root-drenching, control effect of spores in 15 mL fermentation broth was the highest with the efficacy of 70.9%.
