钩状木霉菌的生物学特性及对腐皮镰刀菌的抑菌机理研究

Study on Biological Characteristics of Trichoderma hamatum and the Inhibited Mechanism on Fusarium solani

摘要: 从兰花根际中分离出一株木霉菌株LAN2B-1,对其进行形态学鉴定和ITS序列分析,确定菌株为钩状木霉菌Trichoderma hamatum。该菌株菌落生长的最佳条件为PDA培养基、碳源为甘露醇、pH 5、温度25℃~30℃,暗处理有利于菌落生长,但氮源对菌落生长影响不显著。木霉菌发酵液的抑菌活性和作用机理研究表明,钩状木霉菌发酵液氯仿提取物对腐皮镰刀菌Fusarium solani抑菌活性最佳,生长抑制率为59.38%,其发酵液处理后腐皮镰刀菌菌丝可溶性蛋白和还原糖含量均显著降低。本研究为更好地在生产上利用钩状木霉菌防治由腐皮镰刀菌引起的土传病害奠定了基础。

Abstract: Trichoderma sp. LAN2B-1 isolated from orchid rhizosphere soil was identified as Trichoderma hamatum by morphological method and ITS sequence analysis. T. hamatum grew fastest on PDA medium, with mannitol as carbon source, pH value of 5, temperature of 25℃~30℃ and dark treatment. However, nitrogen source has little effect on its growth. Fermentation broth extracted from T. hamatum by chloroform had antifungal activity against Fusarium solani, with the inhibition rate of 59.38%. Fermentation broth could significantly reduce the soluble protein and sugar content of F. solani. The above results lay a foundation for the better application of T. hamatum against soil-borne diseases caused by F. solani.
