蠋蝽脂肪酶基因的鉴定与表达特征分析

Identification and Expression Profiling of Lipase Genes in Arma custos

摘要: 脂肪酶在昆虫脂质吸收、运输和利用等方面发挥作用。本文采用同源比对方法从蠋蝽Arma custos基因组中鉴定出44个脂肪酶基因,它们分为24个中性脂肪酶、16个酸性脂肪酶、1个GDSL脂肪酶、2个脂肪酶3和1个激素敏感脂肪酶。采用RT-PCR技术分析这些脂肪酶基因在成虫不同组织中的表达特征,结果显示,AcusN3a、AcusN5b、AcusN18c、AcusN22、AcusA13a、AcusLipase3b、AcusGDSL1AcusHSL1A基因在蠋蝽毒液器官(唾液腺)中高表达,AcusN10a、AcusN18b、AcusN20a、AcusN20b、AcusN22、AcusA7a、AcusA7b、AcusA8、AcusA9b、AcusA13a和AcusA15基因在肠道中高表达。对蠋蝽毒液器官各腺体以及肠道中的脂肪酶活性进行测定,发现主腺前叶中的脂肪酶酶活约为23 U/mg protein,主腺后叶中的脂肪酶活性约为21 U/mg protein,肠道中的脂肪酶酶活约为41 U/mg protein。研究结果为后期研究揭示蠋蝽脂肪酶基因在捕食和消化中的生理功能奠定了基础。

Abstract: Lipase plays an important role in lipid absorption, transport, and utilization in insects. In this study, 44 lipase genes were identified in the Arma custos genome by homologous alignment. They are divided into 24 neutral lipases, 16 acid lipases, one GDSL lipase, two lipase 3 and one hormone sensitive lipase. Reverse transcription- polymerase chain reaction (RT-PCR) was used to analyze the expression characteristics of these lipase genes in different tissues of A. custos adults. The results showed that the AcusN3a, AcusN5b, AcusN18c, AcusN22, AcusA13a, AcusLipase3b, AcusGDSL1, and AcusHSL1A genes were highly expressed in the venom apparatus (salivary gland). AcusN10a, AcusN18b, AcusN20a, AcusN20b, AcusN22, AcusA7a, AcusA7b, AcusA8, AcusA9b, AcusA13a, and AcusA15 genes were highly expressed in the gut. The lipase activities in the different glands of venom apparatus and gut of A. custos were assayed. The results showed that the lipase activities in the anterior main gland, posterior main gland, and gut were about 23, 21, and 41 U/mg protein, respectively. These results lay the foundation for further study on the physiological functions of Arma custos lipase genes involved in venom and digestion.
