多黏类芽胞杆菌Pp146的全基因组测序及对药用植物促生防病作用评价

Genome Sequencing of Paenibacillus polymyxa Pp146 and Evaluation of Its Growth-Promoting and Disease Control Effects on Medicinal Plants

摘要: 为系统解析多黏类芽胞杆菌Paenibacillus polymyxa Pp146的促生与生防特性,并评估其在药用植物生产中的应用潜力,本研究结合全基因组测序与平板对峙、盆栽试验开展系统分析。全基因组测序结果显示,菌株Pp146基因组大小为4925561 bp,GC含量为46.84%,共编码4826个蛋白基因,含有14个5S rRNA、13个16S rRNA、13个23S rRNA、132个t RNA、69个ncRNA及9条CRISPRs结构;在NR、Swiss-Prot、COG、GO、KEGG和CAZy数据库中分别注释到4643、3171、4086、3232、2034和238个基因。利用antiSMASH预测到12个次级代谢产物合成基因簇,其中3个与fusaricidin B、tridecaptin及thermoactinoamide A高度同源。与多黏类芽胞杆菌J和SC2的比较基因组分析表明,菌株Pp146基因组较小、GC含量较高,并含有57个特异性基因簇。平板对峙及发酵滤液抑菌试验证实,菌株Pp146对引起地黄、牛膝和山药病害的3种病原真菌抑菌率均超过50.00%,其发酵滤液对孢子萌发最高抑制率达43.38%。盆栽试验,2次预接种菌株Pp146发酵液对3种镰孢菌病害防效最高可达68.94%;单独接种可显著促进宿主植物地下部生物量积累,根鲜质量增加34.24%~164.53%。综上,菌株Pp146兼具广谱抑菌与促生功能,在药用植物生态种植中具有良好开发前景。

Abstract: To systematically elucidate the plant growth-promoting and disease control characteristics of Paenibacillus polymyxa Pp146 and assess its application potential in medicinal plants, this study conducted a systematic analysis combining whole-genome sequencing with plate confrontation and pot experiments. Whole-genome sequencing revealed a genome size of 4925561 bp with a GC content of 46.84%. A total of 4826 protein-coding genes were identified, along with 14 5S rRNA genes, 13 16S rRNA genes, 13 23S rRNA genes, 132 tRNA genes, 69 ncRNA genes, and 9 CRISPRs structures. Additionally, 4643, 3171, 4086, 3232, 2034 and 238 genes were annotated in the NR, Swiss Prot, eggNOG, GO, KEGG and CAZy databases, respectively. Twelve biosynthetic gene clusters(BGCs) for secondary metabolites were identified using antiSMASH, three of which showed high homology with fusaricidin B, tridecaptin, and thermoactinoamide A. Comparative genomic analysis with P. polymyxa J and SC2, which exhibit biocontrol function, revealed that strain Pp146 possesses a smaller genome size, higher GC content, and contains 57 specific gene clusters. Plate confrontation assays and fermentation filtrate inhibition tests confirmed that Pp146 achieved inhibition rates of over50.00% against three pathogenic fungi causing diseases in Rehmannia glutinosa, Achyranthes bidentata, and Dioscorea opposita. Additionally, its fermentation culture filtrate exhibited a maximum spore germination inhibition rate of 43.38%. Pot experiments demonstrated that pre-inoculation with Pp146 culture filtrate twice resulted in a control efficacy up to 68.94% against three Fusarium diseases. Furthermore, single application with Pp146 significantly enhanced the accumulation of belowground biomass in host plants, with fresh root weight increases ranging from 34.24% to 164.53%. In conclusion, strain Pp146 exhibits both broad-spectrum antifungal and plant growth-promoting activities, demonstrating great potential for development in ecological cultivation of medicinal plants.
