饲养温度对蠋蝽胰蛋白酶活性的影响及消化器官形态初步观察

Effect of Temperature on Trypsin Activity and Morphology of Digestive Organs of Arma chinensis (Hemiptera: Pentatomidae)

摘要: 捕食性昆虫的蛋白酶在其消化和生长发育中起着重要作用。为探讨蠋蝽Arma chinensis Fallou环境适应性的消化生理机制,明确蠋蝽在不同季节田间释放的最佳时期,本研究设置15℃、20℃、26℃、30℃和35℃五个饲养温度梯度对蠋蝽成虫的主要消化酶活性进行测定对比。采用双抗体夹心法,形成抗体-抗原-酶标抗体复合物,用酶标仪测定吸光度,依据标准曲线公式计算出蠋蝽唾液腺和中肠胰蛋白酶活性浓度,同时通过使用立体显微镜对蠋蝽分泌消化酶的唾液腺和消化道两个部位及口器的形态结构进行初步观察。结果表明:不同温度下蠋蝽体内胰蛋白酶活性均不相同,并且唾液腺的酶活性均高于中肠的酶活性,胰蛋白酶活性整体上表现出先上升后下降趋势。26℃时唾液腺和中肠中消化酶的活性差异显著,且两部位的酶活性达到最高值,分别为1413.9 IU/L、1029.2 IU/L。形态结构观察表明蠋蝽口器的喙由4个关节组成,蠋蝽唾液腺主要包括一对主唾腺和主唾腺管、一对副唾腺和副唾腺管,整体呈透明状。

Abstract: Proteases play important roles in food digestion and growth and development in predatory insects. The physiological mechanism of digestion and the optimal period for field release in different seasons were investigated through measuring and comparing trypsin enzyme activities of adults of Arma chinensis at five temperatures 15℃, 20℃, 26℃, 30℃, and 35℃. A double antibody sandwich enzyme-linked immunosorbent assay (ELISA) was used to determine the activities of trypsin in the current study. Trypsin activity concentrations in salivary glands and midgut of A. chinensis were calculated according to standard curve formula. Meanwhile, morphology and structure of oral organs, salivary glands and digestive tract of A. chinensis were preliminarily observed. The results showed that the trypsin activity in A. chinensis varied between temperatures, and the enzyme activity in the salivary glands was much higher than that in the midgut. At 26℃, the enzyme activities reached the highest in both the salivary glands and the midgut, which were 1413.9 IU/L and 1029.2 IU/L, respectively, and the difference between them reached the most. The proboscis of A. chinensis consists of four joints. The salivary glands consist of a pair of main salivary glands and a pair of accessory salivary glands.
