Screening of Microorganism Agents for Liriodendron chinense Tissue Culture Seedling and Preliminary Study on Its Mechanism

Abstract: This experiment is aiming to evaluate the impact of using microorganism agent "Nanjing Shield" and 13 strains of arbuscular mycorrhizal fungi (AMF) screened from rhizosphere of tobacco on Liriodendron chinense tissue culture seedling, by detecting the transplanting survival rate and growth-promoting effect. We tried to screen the best microorganism agent for the tissue culture seedling, as well as make clear the basis of its mechanism. The results showed that, the transplanting survival rates of "Nanjing Shield" and GZ38 as high as 99.67%, 96.12% respectively, were significantly higher than the control. Furthermore, the growth-promoting effect of "Nanjing Shield" and GZ38 treatments were 53.71%, 52.84% respectively in greenhouse, and that of GZ38 and "Nanjing Shield" in the field were 26.56%, 18.37% severally. Further experiments showed that GZ38 could colonize at rhizosphere of seedling. GZ38 and "Nanjing Shield" could not only enhance the seedling's root activity and absorption of nutrient elements such as N, P, K, also could improve the contents of invertase, urease, neutral phosphatase and available N, P, K in soil. In addition, GZ38 and "Nanjing Shield" were able to reduce the contents of MDA and improve protective enzymes such as SOD, POD, CAT in seedlings to enhance resistance to adverse environment. Consequently, it is assumed that GZ38 and "Nanjing Shield" might be efficient microorganism agents as the solution of low transplanting survival rate and slowly growth when transplanting Liriodendron chinense tissue culture seedlings to the soil.
