Optimization of Spore-producing Conditions for Lecanicillium cauligalbarum

Abstract: Lecanicillium cauligalbarum is a new species reported in the genus of Lecanicillium, research about it is scarcely. To establish the medium components for culturing this fungus, the optimal carbon source, nitrogen source, and growth factors were screened individually and orthogonal experiments were conducted to screen the best spore-producing conditions. The optimal conditions for sporulation (mass to volume ratio) were:soluble starch 1%, yeast powder 3%, silkworm chrysalis powder 1.5%, potassium chloride 0.05%, dipotassium phosphate 0.1%, magnesium sulfate 0.05%, ferrous sulfate 0.001%, agar 1.5%, distilled water 1000 mL, and temperature of 25℃. L. cauligalbarum was greatly influenced by the nitrogen source but absorbed only low inorganic nitrogen. The spore yield in the optimized medium was 4.5 times of that of PDA and 15.6 times of that of SAB medium. The results provide basic data for the follow-up research on L. cauligalbarum.
