Pathological Changes in Larval Midgut Tissues of Spodoptera litura Feeding on Cry1Ca Protein

Abstract: Cry1Ca has high toxicity to Spodoptera exigua, Spodoptera litura and some other lepidoptera pests. The protein is of great potential to be applied in polygenic strategy of Bt (Bacillus thuringiensis) transgenic crops to enlarge the insecticidal spectrum and manage the insect resistance caused by the current Bt transgenic crop. The toxicity of Cry1Ca to S. litura was studied through observation of the pathological changes of midgut induced by Cry1Ca using transmission electron microscope. The results indicated that the midgut tissues were characterized by obvious pathological changes when the insects were fed Cry1Ca for 2 h, and the pathological changes became ever more obvious with the extension of feeding duration. The main symptoms included swollen, distorted, fractured, and dropped off cell microvilli; stretched or deformed cell nucleus, blurred boundary of karyotheca in cell nucleus, and uneven or gathered chromatins; deformed, cavitated or ridge-blurred mitochondria; and swollen, disordered and reduced endoplasimic reticulum. These results may facilitate the application of Cry1Ca as an important insecticidal protein in the Bt transgenic crops in the future.
