Effects of Transgenic cry30Fa1 Rice on the Expression Pattern of NlHRP in Nilaparvata lugens (Stål)

Abstract: Previous studies found that the transgenic cry30Fa1 rice (KF30-14) could significantly suppress the population growth of brown planthopper (BPH),Nilaparvata lugens (Stål),while the mechanism remains unclear.In this study,using the transcriptome data analysis,a gene called NlHRP with remarkably different expression levels was found and cloned.The result of sequence analysis showed that the NlHRP encoded a histidine-rich protein without conserved domain.RT-qPCR was used to find that the NlHRP expression levels in female BPHs at 24,72 and 96 h after eclosion were higher in KF30-14 experimental population than those in MH86 experimental population,and particularly at 72 h after eclosion were significantly higher.The expression levels of NlHRP in the head,thorax,ovary,midgut and fat body of BPH at 72 h after eclosion were detected by RT-qPCR.The expression levels of NlHRP in the head,midgut and thorax of female BPHs were the highest.The expression level of NlHRP in the head of female BPHs was dramatically increased in KF30-14 experimental population.The results indicate that NlHRP could be related to the BPH adaptation to the transgenic cry30Fa1 rice,and provides the research foundation for the resistant mechanism of transgenic cry30Fa1 rice against BPHs.
