低温冷藏对蠋蝽成虫存活率和主要生化物质的影响

Effect of Low-temperature Storage on Survival Rate and Major Biochemical Substances of Arma chinensis Adult

摘要: 本研究旨在探究低温冷藏对蠋蝽成虫存活率及其体内主要生化物质含量和生化酶活性的影响,以明确蠋蝽在5℃低温保存时长与其生理生化的响应关系。在室内测定低温冷藏时长和不同储存密度对蠋蝽成虫存活率的影响;测定了蠋蝽体内主要能源物质、抗寒物质的含量,以及抗氧化酶的活性变化。研究发现,随着冷藏时间的延长,蠋蝽成虫的存活率逐渐降低,100头/盒为最佳冷藏密度,60 d后存活率高达91%。对于低温胁迫,蠋蝽体内生化物质变化表现出积极的应答机制,成虫显著积累了脂肪、糖原等能源物质和海藻糖、甘油、甘油三酯、山梨醇等抗寒物质,同时海藻糖酶、过氧化物酶和超氧化物歧化酶活性显著增强。随着冷藏时间的延长,这些物质含量和酶活逐渐降低或保持不变。本研究结果对蠋蝽的低温保存及其生理响应机制研究具有重要意义。

Abstract: The aim of this study was to investigate the effects of low temperature storage duration and storage density at 5 ℃ on the survival rate, the contents of main biochemical substances and the activities of enzymes in adult Arma chinensis, in order to determine the relationship between the storage duration and the physiological and biochemical responses of A. chinensis. It was found that the survival rate of adult A. chinensis decreased with prolonged storage. The optimal storage density is 100 per box, with a survival rate as high as 91% after 60 days. In response to the low temperature stress, A. chinensis showed a positive response in biochemical substances. The adults accumulated more energy materials such as fat and glycogen, and cold resistant substances such as trehalose, glycerol, triglyceride and sorbitol. At the same time, the activities of trehalase, peroxidase and superoxide dismutase were significantly enhanced. With the extension of storage duration, the content of these substances and enzyme activity gradually decreased or remained unchanged. The results of this study have great significance for the low temperature storage and physiological response mechanism of A. chinensis.
