多粘类芽胞杆菌NK-1的鉴定、发酵优化及其对采后马铃薯干腐病的生防效果研究

Paenibacillus polymyxa NK-1: Identification, Fermentation Optimization, and Biocontrol Effect on Dry Rot Disease of Postharvest Potato

摘要: 为获得对马铃薯干腐病具有生防潜力的菌株,本研究采用平板对峙法得到一株对Fusarium spp.有良好拮抗活性的菌株NK-1,该菌株对5种病原菌抑菌率为47.31%~91.08%,且该菌株能产生淀粉酶、蛋白酶和纤维素酶;结合形态学观察、生理生化特征及多基因系统发育树分析,鉴定NK-1为多粘类芽胞杆菌Paenibacillus polymyxa;采用单因素试验和正交试验确定NK-1最适发酵成分和条件为蔗糖1%、牛肉膏7%、氯化钠0.1%、硫酸镁0.05%、温度28℃、转速180 r/min、发酵时间48 h、装瓶量50 mL(在250 mL锥形瓶中)、接种量2.5%、初始pH值8,较优化前生物量提高了约60倍,抑菌增长率为4.16%~17.23%;此外,NK-1发酵液可导致病原菌菌丝畸形、消解,浓度为108 CFU/mL时孢子萌发率为16.17%~24.50%;马铃薯贮藏试验结果表明,第30 d时NK-1发酵液处理对马铃薯干腐病的预防和防治效果分别为30.10%和23.01%。表明菌株NK-1对马铃薯采后干腐病具有较好的生防潜力,为干腐病生物防治提供了新菌种资源。

Abstract: In order to obtain strains with potential biocontrol ability against potato dry rot, NK-1, a strain with remarkable antimicrobial activity against Fusarium spp., was isolated using the plate confrontation assay in the present study. Strain NK-1 demonstrated antagonism against five pathogens, with inhibition rates of 47.31%-91.08%, and produced amylase, protease, and cellulase. Combined with morphological observations, physiological and biochemical characteristics, and multi-gene phylogenetic tree analysis, NK-1 was identified as Paenibacillus polymyxa. Through single-factor and orthogonal tests, the optimal fermentation components and conditions for NK-1 were determined as: 1% sucrose, 7% beef extract, 0.1% NaCl, 0.05% MgSO4, temperature 28 ℃ , agitation speed 180 r/min, fermentation time 48 h, culture volume 50 mL (in 250 mL conical flasks), inoculum concentration 2.5%, and initial pH 8. Under these conditions, the biomass of NK-1 was approximately 60 times more than that obtained under pre-optimisation conditions. Additionally, its antimicrobial activity growth rate ranged from 4.16% — 17.23%. Moreover, the fermentation broth of NK-1 could induce deformation and disintegration of pathogenic hyphae. The germination rate of pathogenic fungal spores treated with 108 CFU/mL of NK-1 fermentation broth ranged from 16.17%—24.50%. Results of the potato storage experiment showed that the NK-1 fermentation broth exhibited significant efficacy in controlling potato dry rot. The preventive and therapeutic effects were 30.10% and 23.01%, respectively, by day 30. This demonstrates that strain NK-1 possesses promising biocontrol potential against potato post-harvest dry rot, providing a new microbial resource for biological control of Fusarium dry rot.
