九龙山自然保护区四种生境土壤的虫生真菌多样性

Entomogenous Fungus Diversity of Four Habitats in Jiulong Mountain Nature Reserve

摘要: 为明确浙江省九龙山国家级自然保护区不同生境下的土壤虫生真菌多样性及分布特点,给该区域虫生真菌的开发利用及物种多样性提供科学依据,本研究以九龙山自然保护区四种不同生境(茶园、红薯地、人工林、原始林)的土壤为研究材料,通过illumina NovaSeq测序平台测序得到有效数据,分析四种生境下20个土壤样品中的真菌多样性与虫生真菌的群落结构组成,探讨虫生真菌与土壤环境因子的相关性。结果发现:四种生境下林地(原始林、人工林)土壤的虫生真菌相对丰度高于农业用地(茶园、红薯地);四种生境土壤的虫生真菌群落具有显著差异,原始林与人工林中的最优势属是弯颈霉属,茶园与红薯地中的最优势属分别为镰刀菌属和绿僵菌属;该地区土壤碳氮比和全氮水平是影响虫生真菌菌群的极显著环境因子,林地土壤的虫生真菌菌群与土壤碳氮比、全氮水平正相关,农业用地土壤中的虫生真菌群落与土壤全磷、速效磷含量正相关;四种生境土壤虫生真菌相对丰度排行前十的属中,绿僵菌属、棒束孢属、虫草属与土壤碳氮比、全氮水平极显著正相关,镰刀菌属与其极显著负相关。该研究结果表明,九龙山自然保护区虫生真菌种类丰富,生境类型影响虫生真菌的丰富度,林地土壤的虫生真菌相对丰度较高。该地区的优势虫生真菌主要集中于3个属:绿僵菌属、弯颈霉属、镰刀菌属。

Abstract: In this study, soil samples from four different habitats (tea garden, sweet potato field, plantation and primary forest) in Jiulongshan National Nature Reserve of ZheJiang Province were used as research materials. Effective data were obtained by illumina NovaSeq sequencing platform for analysis of the fungal diversity and community structure of entomogenous fungi in 20 soil samples under four different habitats, and the correlation between entomogenous fungus and soil environmental factors, The main purpose was to clarify the diversity and distribution characteristics of soil entomogenous fungus in different habitats and provide scientific basis for the development and utilization of entomogenous fungus and species diversity in Jiulongshan National Nature Reserve, Zhejiang Province. The results showed that the relative abundance of entomogenous fungus in forests (primary forest, plantation) was higher than that in agricultural land (tea garden, sweet potato field) under four habitats. There was a significant difference in the entomogenous fungus communities in the soils of the four habitats. The dominant genera in the primary forest and plantation were Tolypocladiumi, while those in the tea garden and sweet potato field were Fusarium and Metarhizium respectively. The ratio of carbon to nitrogen and total nitrogen in soils were significant environmental factors affecting the entomogenous fungus in the region. In forest soils, entomogenous fungus floras was positively correlated with soil carbon to nitrogen ratio and total nitrogen level, while in agricultural soils, the contents of total phosphorus and available phosphorus were positively correlated with entomogenous fungus communities. Among the top ten genera in the relative abundances of soil entomogenous fungi in four habitats, Metarhizium, Isaria and Cordyceps were significantly positively correlated with soil carbon to nitrogen ratio and total nitrogen level, while Fusarium was significantly negatively correlated with soil carbon to nitrogen ratio. The results showed that there were rich entomogenous fungi species in Jiulongshan Nature Reserve, habitat type influenced the abundance of entomogenous fungi, and the relative abundance of entomogenous fungi in the soils of forest was higher than that in the four different habitats. The dominant entomogenous fungi mainly concentrated in three genera, such as Metarhizium, Tolypocladium and Fusarium.
