Screening, Identification of Streptomyces yangpuensis WLU210 and Its Biocontrol Effect on Fusarium Wilt of Bitter Gourd

Abstract: Soil samples were collected from Yellow River wetland at Henan. A dilution coating flat plate method and dual culture assay were applied to isolate actinomycetes strains and evaluate their antimicrobial activity. Finally, one actinomycete strain WLU210 with stable antimicrobial activity were obtained. Based on morphological, physiological and biochemical characteristics along with 16S rRNA gene sequence analysis, the strain WLU210 was identified as Streptomyces yangpuensis. Both S. yangpuensis WLU210 strain and its extract exhibited a broad-spectrum antagonistic activity against seven plant pathogens, such as Fusarium oxysporum f. sp. momordicae, Botryosphaeria dothidea, Fusarium pseudograminearum, Phytophthora capsici, Colletotrichum orbiculare, Gaeumannnomyces graminis var. tritici and Setosphaeria turcica. In addition, the fermentation broth of S. yangpuensis WLU210 could promote the growth of bitter gourd in pot and field tests. Compared to the chemical agent carbendazim with the field control efficacy of 73.89% against bitter gourd Fusarium wilt, strain WLU210 displayed equal control efficacy of 68.89% by root-drenching with 106cfu/mL spore suspension. In summary, S. yangpuensis WLU210 has the potential to be developed into a biocontrol agent for bitter gourd Fusarium wilt control.
