Function Analysis of LmNotch in the Wing Development of Locusta migratoria

Abstract: Notch is a receptor protein in the Notch signaling pathway,regulating the expression of downstream target genes after binding with ligands and playing an important role in the development and differentiation of invertebrates and vertebrates.In this study,the Notch signaling receptor gene LmNotch was identified based on transcriptome database of Locusta migratoria,and the expression characteristics of LmNotch in different tissues and wing pads of different development days of the fifth-instars were analyzed by qRT-PCR.The biological function of LmNotch was analyzed by RNA interference experiment.Compared with the control group,70% of the locusts injected with dsLmNotch could not complete the molting process,and 10% of the locusts showed wing disorder phenotype after molting.The results of H&E staining on the paraffin section of wing pads showed that,compared with the control group,the cavity space between the upper and lower cuticles of the wing in the treatment group was enlarged,and a complete new cuticle could not be formed.The expression of other wing developing-related genes,such as Dpp,Omb,Yorkie,and the wing specific cuticle protein genes,was affected by the decrease of LmNotch expression.These results shed light on the mechanism of insect wing development and provide potential targets for pest biological control.
