Isolation and Identification of Plant Endophyte Bacterial Bacillus tequilens and Its Control Effect against Watermelon Fusarium Wilt

Abstract: For the purposes of selecting new biocontrol agents, bacteria wm031 was isolated from the tomato plants using the dilution plate method and its antifungal activity was tested by the dual plate assay. The strain wm031 was identified based on morphological characteristics, 16S rDNA sequence analysis, physiological and biochemical characteristics, and its antagonistic against plant diseases was investigated by greenhouse experiments. The results indicated that the strain wm031 exhibited broad-spectrum antagonism against the tested fungi, including Fusarium oxyporum f. sp. niveum, Sclerotinia sclerotiorum in oilseed rape, Bipolaris sorokinianum, Gaeumannomyces graminis var. tritici Walker, Phytophthora capsici, Sclerotinia allii Saw., Fusarium moniliforme Sheld, Colletotrichum gloeosporioides, and Fusarium oxysporum f. sp. lycopersici. The strain wm031 was identified as Bacillus tequilens, and it could stably colonize in watermelon and rice plants. Strain wm031 demonstrated good control effects on watermelon Fusarium wilt caused by Fusarium oxyporum f. sp. niveum, with the efficacy of 74.6%, higher than that of chlorothalonil by 14.1%. Thus, the strain wm031 is a potential biocontrol agent to be applied in the field.
