Isolation and Identification for Arthrobacter nicotinovorans against Locust from Qinghai-Tibetan Plateau and Optimization of Culture Conditions

Abstract: A bacteria strain selected from the soil of Qinghai-Tibetan Plateau was identified as Arthrobacter nicotinovorans and named GZ1-6 based on the 16S rRNA sequence analysis, physiological and biochemical characteristics. Virulence of GZ1-6 to nymph of Locusta migratoria manilensis was evaluated in the laboratory, with a corrected accumulative mortality over 75.56%. The optimum fermentation conditions were determined to be temperature 35℃, pH 7.5, sucrose and yeast extract as carbon and nitrogen source, respectively. The optimized fermentation products showed a 7.7% higher toxicity against L. migratoria manilensis compared with pre-optimum. The results indicate that the strain GZ1-6 can be used as a potential biocontrol agent against the locust in the field.
