Optimization of Solid-state Fermentation Process of Paecilomyces lilacinus M-1 and Pilot Production based on Automatic Seed Koji Machine

Abstract: Paecilomyces lilacinus has a great potential in biological control of the plant nematodes, however, the immature technology of mass production and short shelf life limit its industrial production. Thus, in order to obtain the optimum conditions for solid fermentation of P. lilacinus M-1 and to establish its industrial production technology, single factor experiment and orthogonal experiment were used in this study to optimize its fermentation conditions, such as culture medium composition, fermentation conditions and drying conditions. The results showed that the optimized composition for solid fermentation media of P. lilacinus M-1 was as followed:bran:corn (1:1), sucrose addition (5%), urea addition (0.1%), ammonium sulfate (0.1%) and the ratio of material to water (1:0.67). The optimum culture conditions of solid fermentation was as followed:culture temperature (28℃), culture time (7 d), liquid inoculation amount (5%) and solid inoculation amount (0.5%). The optimum drying condition of the fungal inoculum was 35℃ for 24 h. Under this optimal condition, the number of living cells in P. lilacinus M-1 solid inoculant was 9.47×109 CFU/g. Then, on this basis, the large-scale solid fermentation process of P. lilacinus M-1 was established by means of automatic seed koji, and verified by 3 batches of larger-scale production, nearly 2 t P. lilacinus M-1 solid inoculant were obtained, which had 15.6×109 CFU/g living cells and had a very low rate of miscellaneous bacteria (<0.01%), and the moisture content was 9.68%. In conclusion, this process can be used in the industrial production of P. lilacinus M-1 and the product quality is excellent.
