一株罹病草地贪夜蛾成虫莱氏绿僵菌的分离鉴定及其致病力

Isolation, Identification and the Virulence of Metarhizium rileyi from Adult of Spodoptera frugiperda (J. E. Smith)

摘要: 从草地贪夜蛾成虫尸体中分离获得一株杀虫真菌。经形态和分子鉴定,确定该虫生真菌为莱氏绿僵菌,并命名为NrSfadult202104。生物学特性的研究结果显示,该菌株在SMAY+0.5%草地贪夜蛾蛹粉培养基上生长速率和分生孢子产量最佳;在28 ℃时菌落生长速率最快,20 ℃时产孢最为适宜;在全光照条件下该菌株产孢量最高。对草地贪夜蛾成虫和3龄幼虫的致病力测定结果显示,该菌株对草地贪夜蛾雌、雄成虫以及3龄幼虫都具有一定的毒力效果。当孢子浓度为1.0×108孢子/mL时,其感染雌、雄成虫校正死亡率分别为61.2%和58.1%;菌株接种3龄幼虫后,随孢子悬浮液浓度的升高,幼虫的感病死亡率也随之增加,当浓度达到1.0×108孢子/mL时,3龄幼虫的致死中时(LT50)为5.30 d,接种8 d后3龄幼虫的校正死亡率为66.12%。综合以上结果,莱氏绿僵菌NrSfadult202104菌株对草地贪夜蛾具有潜在的生防作用,值得深入研究。

Abstract: A strain of entomopathogenic fungi was isolated from the adult of Spodoptera frugiperda. On the base of morphological and molecular analyses, this strain was identified as Metarhizium rileyi and named NrSfadult202104. In this study, the phenotypic characteristics of the strain were evaluated. The maximal growth rate of colony and conidial production were on the SMAY medium containing puparium powder. The optimal temperatures for colony growth and conidial production were 28 ℃ and 20 ℃, respectively. The maximum conidial production was achieved under the 24h-illumination condition. This strain was pathogenic to adults and the 3rd instar larvae of S. frugiperda, the corrected mortality of infected female and male adults were 61.2% and 58.1%, respectively with the inoculation of conidial concentration of 1.0×108 spores/mL. The mortality of 3rdinstar larvae increased with the increase of the conidial concentration, the median lethal time (LT50) of the larvae was 5.30 days with the concentration of 1.0×108 spores/mL, and the corrected mortality reached 66.12% on the 8th day. In summary, the strain NrSfadult202104 shows a good potential as a biocontrol agent for S. frugiperda and is worth of further research.
