虎杖提取物对苹果腐烂病菌的抑菌机制

Mechanism of Extracts Obtained from Polygonum cuspidatum to Suppress Valsa mali

摘要: 本文采用乙醇热回流法提取虎杖,以苹果腐烂病菌为指示菌,利用固体培养方法研究虎杖乙醇提取物对病菌的抑制率,通过液体培养方法研究其对菌丝形态、菌丝干重等的影响,同时探究其对菌丝体内几丁质酶活性、β-1,3-葡聚糖酶活性、蛋白酶活性、可溶性蛋白含量及还原糖含量等一些生理生化指标的影响。结果表明,虎杖乙醇提取物对苹果腐烂病菌有明显的抑制作用。当虎杖提取物浓度达800 mg/L时,对菌丝干重的抑制率高达96.5%;抑制蛋白、葡萄糖等菌体细胞内物质的合成,其含量最高可降低为对照的45.2%和23.0%,从而使病菌代谢速度减慢,抑制其生长;使2种细胞壁相关水解酶,几丁质酶和β-1,3葡聚糖酶的活性分别升高3.36和7.98倍,降解细胞壁,破坏菌体结构,使菌体自溶。

Abstract: The ethanol extract from Polygonum cuspidatum Sieb. et Zucc. by hot reflux extraction inhibited radial growth to Valsa mali Migable et Yamada in solid culture systems. Liquid culture tests showed that the extracts at 800 mg/L inhibited mycelium dry weight by 96.5%. The synthesis of protein and glucose was inhibited to 45.2% and 23.0% of the control, respectively. Activities of chitinase and β-1,3-glucanase was 3.36 and 7.98 times higher than those of control, favorable to suppression of pathogenic fungi.
