哈茨木霉FJAT-9040生防菌剂固体发酵及其对苦瓜枯萎病的防治效果

Solid-state Fermentation of Biocontrol Agent Trichoderma harzianum FJAT-9040 and Its Efficiency against Bitter Gourd Fusarium Wilt

摘要: 为优化枯萎病生防菌哈茨木霉FJAT-9040生防菌剂的固体发酵条件,本研究以农业副产物--微生物发酵床养猪垫料为主要培养基成分,采用聚丙烯塑料袋培养法进行菌株FJAT-9040的固体发酵,分析了接种方式、装料量和培养时间等因子对该菌株生长的影响,并测定了发酵产物对苦瓜植株生长的影响及其对苦瓜枯萎病的田间小区防治效果。结果表明,拌菌后装袋培养处理组优于装袋后接菌培养处理组。综合比较发现,采用拌菌后装袋培养,当装料量为60%~70%时,第18~20 d时产孢量达到最大值,为(4.33~4.46)×108孢子/g,较初始接菌量增加了4个数量级。此时,外观观察发现菌丝体呈深绿色,且长满整个培养袋。使用5%和10%哈茨木霉FJAT-9040生防菌剂处理苦瓜苗,能显著促进植株根系与藤蔓的增长,比对照组分别长(8.0~8.3)mm和(27.5~51.2)mm,但对叶片数影响不显著。移栽时及移栽后共使用2次该生防菌剂对田间苦瓜枯萎病防治效果为61.21%,与相同处理方式的化学药剂多菌灵的防治效果59.93%相当。

Abstract: To optimize solid-state fermentation conditions for biocontrol agent Trichoderma harzianum FJAT-9040, the agricultural by-product pig-raising beddings fermented by microbes were used as the main medium components and the polypropylene plastic bagged cultivation method was used to perform the solid-state fermentation of the FJAT-9040 strain. The effects of inoculation method, medium volume and culturing time on growth of the FJAT-9040 strain were analyzed comprehensively. Subsequently, the effects of the fermentation products on bitter gourd growth and their biocontrol efficiency against bitter gourd Fusarium wilt in field plot experiments were detected, respectively. Our results demonstrated that the treatment groups of inoculation before compost bagging were superior to those of inoculation after compost bagging in the aspect of inoculation method. The results of comprehensive comparison showed that colonies yield of the FJAT-9040 strain reached its maximum on the 18th to 20th day when the medium volume was 60%-70% and the inoculation method was inoculation before compost bagging. The highest colonies yield of the fermentation products could be (4.33- 4.46)×108 spores/g, which was increased four orders of magnitude in relative to the initial inoculum dosage. At this time, the hyphae of FJAT-9040 were bottle green according to the observation of the appearance and broadly distributed the whole culture bag. Importantly, the treatment of bitter gourd seedlings with 5% or 10% T. harzianum FJAT-9040 biological fertilizers could significantly promote the growth of root length with increase of (8.0-8.3) mm and vine length with increase of (27.5-51.2) mm, whereas the treatment had no significant influence on leaf number. After the twice treatments with T. harzianum FJAT-9040 biological fertilizers, the control efficiency against bitter gourd Fusarium wilt could be 61.21%, which was very similar to the control efficiency (59.93%) of treatment with the chemical agent carbendazol using the same treatment methods.
