青枯劳尔氏菌蛋白PopW表达条件优化及其诱导植物抗病功能

Induction of Protein PopW from Ralstonia solanacearum in Plants Disease Resistance

摘要: 蛋白PopW是从青枯劳尔氏菌ZJ3721中鉴定得到的一种新的Harpin蛋白,为降低成本,我们对诱导剂IPTG的浓度进行了筛选,结果表明,诱导PopW蛋白表达的最适IPTG浓度为0.01 mmol/L。对蛋白PopW在诱导烟草抗TMV防卫反应中最适宜浓度和持效性的研究结果表明,用蛋白PopW处理的烟草在接种TMV后,其叶片上的枯斑数显著少于对照,且PopW浓度为2.5 mg/L时持效性可达8 d。同时,用蛋白PopW预处理的烟草接种TMV后,其叶片中PR-1a的表达量显著高于对照。浓度为2.5 mg/L的蛋白PopW可以分别诱导番茄对叶霉病、水稻对稻曲病的抗病性,防效达80.19%和86.84%。蛋白PopW对辣椒还有一定的促生作用,田间经2.5 mg/L的PopW蛋白处理后可增产16.89%。

Abstract: A new extracellular harpin protein PopW was identified from Ralstonia solanacearum ZJ3721. The expression of recombinant protein PopW could be induced by IPTG, and 0.01 mmol/L was demonstrated to be an optimal concentration for its peak expression. There were significantly less diseased lesions in tobacco leaves treated with PopW followed by the inoculation of TMV than that of control plants, and this induced resistance could last more than eight days with 2.5 mg/L PopW. Moreover, the expression of PR-1a was significantly higher in tobacco leaves treated with PopW than that of control leaves. PopW was also demonstrated to control tomato leaf mold and rice false smut in field with efficiency of 80.19% and 86.84%, respectively. When treated with 2.5 mg/L PopW, pepper planted in field obtained a good yield with an increase of 16.89% compared with the control.
