南海深海甲基营养型芽孢杆菌SHB114抗真菌脂肽活性产物的研究

Anti-fungal Lipopeptides Produced by Bacillus methylotrophicus SHB114Isolated from South China Sea

摘要: 南海深海来源细菌SHB114具有抗真菌活性,经鉴定为甲基营养型芽孢杆菌Bacillus methylotrophicus,为首次报道从海洋中分离得到。该菌摇瓶发酵液经酸沉淀、有机溶液抽提、快速硅胶柱色谱、中低压制备液相色谱与半制备HPLC逐级分离纯化,活性追踪抗丝状真菌的活性物质,分离纯化得到3个脂肽类化合物。采用核磁共振光谱与质谱将活性化合物鉴定为bacillomycin Lc类脂肽。其具有较强抑制黄瓜炭疽病菌、立枯丝核菌、黄瓜枯萎病菌等植物病原真菌的活性,有防治作物真菌病害的研发价值。

Abstract: Bacillus methylotrophicu SHB114 was firstly isolated from deep-sea sediment collected from South China Sea. Three anti-fungal lipopeptides of the bacillomycin Lc family were purified from the fermentation broth via bioactivity-guided fractionation and isolation based on acid precipitation, vacuum flashchromatography, and semi-preparative HPLC. The three compounds were identified by interpretation of their NMR and MS spectroscopic data. All compounds showed significant inhibitory activities against fungal plant pathogens including Colletotrichum orbiculare, Rhizoctonia solani and Fusarium oxysporum f. sp. cucumerinum in paper-agar disk diffusion assay.
