Inhibition of Bacillus velezensis HN-2 Fermentation Supernatant on Three Kinds of Xanthomonas

Abstract: Bacillus velezensis strain HN-2 is a biocontrol bacterial strain isolated from soil in our laboratory. In this study, we investigated the optimal extraction methods, inhibition activity and stability of active components in the fermentation supernatant of strain HN-2. The results showed thatn-butyl alcohol extract, ammonium sulfate precipitation extract and acidification extract of B. velezensis HN-2 exhibited potent inhibitory activity against three kinds of Xanthomonas. The n-butyl alcohol extract had the strongest activity against Xanthomonas oryzae pv. oryzicola with the inhibition zone of (41.57±0.33) mm. Compared with bacitracin, kasugamycin and bismerthiazol, the n-butyl alcohol extract at 10 mg/mL had higher antibacterial activity. Stability experiment showed that the active substance of n-butyl alcohol extract had a broad adaption range to pH value (pH 1.0-9.0), and was stable under high temperatures and UV-treatment. In conclusion, the results revealed that the n-butyl alcohol extract of strain HN-2 with strong antibacterial activity and stability had the potential to be developed into biocontrol agent.
