Identification and Biological Characteristics of a Metarhizium guizhouense Strain Isolated from Blattaria germanica

Abstract: In this study, a Metarhizium strain RCEF6416 was isolated from naturally diseased Blattaria germanica adults collected from Magu mountains forestry center, Xuancheng, Anhui Province. The pathogen was identified as Metarhizium guizhouense by morphological and molecular biological methods. To provide strain resource and theoretical basis for biological control of the important health pest B. germanica, we investigated the culture growth and sporulation ability under different conditions of culture medium, salt concentration, pH value, and high temperature stress. The results showed that the strain RCEF6416 grew the fastest on PPDA medium, with daily increase of colony diameter of 4.94 mm/d. However, the strain sporulated the highest on PDA medium, reaching 5.1×107 spores/cm2. In a certain range of salt concentration, the daily increase of colony diameter increased with the increase of salt concentration, whereas the spore production decreased gradually, and the colony produced white hyphae circle. The optimum pH for culture growth and sporulation capacity was 8. High temperature inhibited the spore germination rate. When treated for 4 h at water bath of 45℃, the spore germinated at only 2.6%.
