Optimization of Solid-State Fermentation Process of Penicillium simplicissimum CEF-818

Abstract: The fermentation product of Penicillium simplicissimum CEF-818 can effectively control cotton Verticillium wilt caused by Verticillium dahliae. In order to optimize fermentation conditions of P. simplicissimum CEF-818, the composition of solid fermentation medium and fermentation conditions of CEF-818 were screened by single factor and orthogonal test. The results showed that the optimum culture for solid fermentation of P. simplicissimum CEF-818 was wheat bran, and addition of 2.5% glucose (M/M), 0.6% urea (M/M) and 0.6% potassium chloride (M/M), with the initial pH value of 7.0, the ratio of material to water 1:0.5, the liquid inoculums level of 5% (V/M), the material thickness of 2 cm and at 28℃ for 7 days. Under this optimal medium and fermentation process, the sporulation could reach to 8.28×109 CFU/g for P. simplicissimum CEF-818.
