Pathological Changes in Larval Midgut Tissues of Ostrinia furnacalis (Lepidoptera:Crambidae) and Mythimna separata (Lepidoptera:Noctuidae) after Feeding Cry1F Protein

Abstract: To determine the action mechanism of the insecticidal protein Cry1F, the pathological changes in the midgut of the Asian corn borer, Ostrinia furnacalis (Guené) and Oriental armyworm, Mythimna separata (Walker), fed on an artificial diet containing Cry1F protein, were observed using transmission electron microscope, and the changes induced by Cry1Ab were also investigated. The results showed that the goblet cells changed significantly after Asian corn borer and Oriental armyworm fed on an artificial diet containing Cry1F protein for 48 h, the major symptoms included:microvilli swelled and dropped off, the nucleus deformation, the boundary of karyotheca in cell nucleus blurred and the chromatins underwent pyknosis and was pressed close to the karyotheca, the number of mitochondria tensile deformation and endoplasmic reticulum reduced, the inner cristae of mitochondria indistinct and vacuolated, and the endoplasimic reticulum were swollen, fractured and disordered. The pathological changes of goblet cells in the midgut of Asian corn borer and Oriental armyworm larvae induced by Cry1F were the similar to that by Cry1Ab. This study may provide the theoretical basis for better using Cry1F as important insecticidal protein in transgenic corn in the future.
