Identification of an Endophytic Fungus of Podocarpus macrophyllus and Its Antifungal Metabolites

Abstract: The endophytic fungal strain ZJDH255 with antifungal activity was isolated from Podocarpus macrophyllus in Jiande of Zhejiang, and was identified as Nigrospora oryzae based on its morphological characters and ITS rDNA sequence. Bioassay showed that the crude extract of the ZJDH255 fermentation broth exhibited strong inhibitory activities against plant pathogenic fungi Botrytis cinerea and Colletotrichum orbiculare. Three antifungal compounds were separated from the crude extract through normal-phase silica gel column chromatography and Sephadex LH-20 column chromatography. They were identified as mellein, griseofulvin and dechlorogriseofulvin based on mass spectrometry and nuclear magnetic resonance spectroscopy.
