Optimization of Fermentation Conditions for Erythrobacter sp. YH-07 Suppressing Fusarium Wilt of Tomato

Abstract: A marine bacteria Erythrobacter sp. YH-07 with high biocontrol activity against Fusarium oxysporum was isolated from Lianyungang beach. We used single factor experiments and orthogonal experiments to investigate the best condition for fermentation of strain YH-07. The best composition of the medium was 0.25% (W/V) yeast extract, 1% (W/V) source, 1% (W/V) tryptone, 3% (W/V) NaCl and 0.03% (W/V) MgSO4·7H2O. The optimal fermentation conditions were the combination of temperature 30℃, initial pH7.0, 140 r/min, 1% inoculums volume percentage and medium volume 50 mL/250 mL. Under the optimum fermentation medium and culture conditions, the inhibitory rate of strain YH-07 was 9.4% higher than before. Pot experiments indicated that H7+R treatment could significantly lower the incidence rate of Fusarium wilt disease up to 50%, increase the total number of bacteria and actinomycetes, and decrease the number of fungi in rhizosphere soil. Strain YH-07 can effectively control Fusarium wilt of tomato and improve microbial community structure in rhizosphere, and it has potential development and application prospect.
