血根碱对菜青虫几种代谢酶活性的影响

Effect of Sanguinarine on Metabolic Enzymes of Pieris rapae

摘要: 血根碱是博落回Macleaya cordata (Willd.) R. Br.的主要杀虫活性成分之一。为探讨其作用机理,以叶碟饲喂法处理菜青虫Pieris rapae L. 3龄幼虫,测定了处理后试虫体内几种重要酶系的活性。结果表明:血根碱对菜青虫羧酸酯酶、碱性磷酸酯酶、酸性磷酸酯酶和细胞色素P450有明显的激活作用;对谷胱甘肽S-转移酶的影响较为复杂;对乙酰胆碱酯酶具有强烈的抑制作用。其中羧酸酯酶处理后36 h比活力最大,为同期对照的2.20倍;碱性和酸性磷酸酯酶分别于处理后12 h和24 h逐渐被激活,且与对照差异显著;细胞色素P450处理后48 h比活力为同期对照的3.70倍;对乙酰胆碱酯酶36 h抑制率最大为95.12%。可见,血根碱可明显影响昆虫的代谢酶系,且其对乙酰胆碱酯酶抑制作用可能与其毒杀作用有关。

Abstract: Sanguinarine is one of the main insecticidal ingredients extracted from Macleaya cordata. To elucidate the insecticidal mechanism, the effects of sanguinarine on six metabolic enzymes of 3rd instar larvae of Pieris rapae were measured. The results showed that the activities of carboxyl esterase (CarE), alkaline phosphatase (ALP), acid phosphoesterase (ACP) and cytochrome P450 in treated larvae were distinctly activated by sanguinarine, while the activity of acetylcholinesterase (AChE) was obviously inhibited. Effect on activity of glutathione S-transferases (GSTs) was complex. The specific activity of CarE was 2.20 times higher than that of the control at 36 h after treatment; the activities of ALP and ACP were the highest at 12 and 24 h after sanguinarine treatment, respectively. The specific activity of cytochrome P450 in treated insects was increased 3.70 times at 48 h after treatment. The activity of AChE was inhibited the most at 36 h after treatment by 95.12%. Obviously, sanguinearine affects the metabolic enzymes, and the inhibition of AChE may be related with its lethal activity.
