不同施肥处理对土壤微生物群落结构及西瓜枯萎病发生的影响

Effects of Different Fertilization Treatments on Soil Microbial Community Structure and the Occurrence of Watermelon Wilt

摘要: 研究不同肥料组合对西瓜枯萎病发生的影响,寻找与西瓜幼苗发病密切相关的菌群种类。设置3组不同土壤肥料组合,依次为对照组CK、T1、T2。利用Illumina Miseq高通量测序平台对试验组和对照组之间的土壤微生物群落组成在16S rDNA水平上进行测序分析。通过对测序结果在OTU和种群相对丰度的计算和分析发现,CK组中平均共获得28986条序列,T1组中平均共有30308条序列,T2组中平均共有29730个条序列;三组共有细菌OTU数量为860,占各组分平均总OTU数量的50%以上,其中芽胞杆菌属Bacillus、肠球菌属Enterococcus、乳球菌属Lactococcus、梭菌属Clostridium在T1组中相对丰度最高;而2种未知细菌unclassified_TM7-1(c)unclassified_Ellin329(o)在T2组中的相对丰度值最高。西瓜枯萎病发病率情况为CK>T1>T2,施用棉隆+生物有机肥可以通过重建土壤生态环境,提高土壤细菌多样性,从而对西瓜枯萎病有显著的防治作用。

Abstract: The objective of this study was to compare the effects of different fertilizer combinations on occurrence of fusarium wilt of watermelon and to excavate the closely related genus to the disease of watermelon seedlings. The composition of bacterial communities of soil samples fertilized by 3 different soil fertilizer combination groups, followed by the control group (CK), T1 and T2, were compared to that of bacterial communities by the Illumina Meseq platform targeting the V3-V4 regions of 16S rDNA. Through the sequencing results analysis in OTU and the relative abundance of the population, we have find 28986 tags in CK, 30308 tags in T1, and 29730 tags in T2, and there are 860 OTUs in common, which is more than 50% within average of each treatment. And in the genus top 10 bacteria population, the abundance of Bacillus, Enterococcus, Lactococcus, Clostridium in T1 was apparently higher than others, while the unclassified_TM7-1(c) and unclassified_Ellin329(o) were significantly high in T2. The incidence of watermelon Fusarium wilt is CK > T1 > T2. The application of Basamid+Bio-organic fertilizer can be used to rebuild the soil ecological environment and help to improve the soil bacterial diversity, which has a significant preventive effect on watermelon fasurium wilt.
