Optimization of Solid Substrate and Conditions of Trichoderma asperellum DQ-1 and Effect on Four Crop Seedlings Growth

Abstract: In this study, the solid fermentation substrate and conditions of Trichoderma asperellum DQ-1 were screened and optimized by single factor and orthogonal test using wheat bran, corn flour, rice husk, straw and bagasse as raw materials. The effects of fermentation products on the growth of cucumber, pepper, tomato and watermelon seedlings were investigated by pot experiment. Single factor tests showed that T. asperellum DQ-1 with single wheat bran as the fermentation substrate could produce the largest amount of conidia of, and the amount of conidia was 1.51×109 CFU/g after 5 days of fermentation. When wheat bran, corn flour, rice husk and diatomaceous earth were combined at the proportion of 5.8:0.5:1.7:1, the conidia yield of T. asperellum DQ-1 reached 2.63×109 CFU/g after 5 days of fermentation. Under the condition of solid fermentation with liquid inoculation at the inoculation amount of 2%, the conidia yield of T. asperellum DQ-1 was the highest, up to 7.18×109 CFU/g, on the 12th day of fermentation. The pot experiment demonstrated that the T. asperellum DQ-1 conidia powder had obvious growth promotion effect on the seedlings of cucumber, pepper, tomato and watermelon. The above results will lay a foundation for the industrial production and application of T. asperellum DQ-1.
