Identification and Characterization of Burkholderia cepacia JX-1 against the Tomato Bacterial Wilt

Abstract: Strain JX-1, which was isolated form the rhizosphere soil, exhibited high levels of biocontrol efficacy against tomato bacterial wilt caused by Ralstonia solanacearum under greenhouse conditions. Strain JX-1 was identified as Burkholderia cepacia based on morphological, physio-biochemistry features, and sequence analysis of 16S rDNA and gyrB. Strain JX-1 produces several antimicrobial compounds, such as siderophores and extracellular proteinase. PCR analysis demonstrated that the specific genes within the biosynthetic loci for pyrrolnitrin and pyoluterin are conserved in strain JX-1. Random mutagenesis was performed to identify novel regulator of antimicrobial traits in strain JX-1, and the mutants with significantly increased or decreased the antimicrobial activity were identified. Our data showed that the antimicrobial activity was negatively regulated by gntR gene, and positively influenced by the tlpks/nrps gene in strain JX-1. Our findings indicated that JX-1 could produce multiple secondary metabolites and the tlpks/nrps and gntR genes played a vital role on its biocontrol capacity.
