杨浦链霉菌WLU210的筛选、鉴定及其对苦瓜枯萎病的生防作用研究

Screening, Identification of Streptomyces yangpuensis WLU210 and Its Biocontrol Effect on Fusarium Wilt of Bitter Gourd

摘要: 本研究从河南黄河湿地采集土壤样品,采用梯度稀释涂布平板法分离和平板对峙培养法筛选获得一株对苦瓜枯萎病菌具有稳定拮抗活性的链霉菌WLU210。在形态学鉴定和生理生化特征测定的基础上,对菌株WLU210的16S rRNA基因序列进行比对分析,将WLU210鉴定为杨浦链霉菌Streptomyces yangpuensis;拮抗谱测定结果表明,杨浦链霉菌WLU210对苦瓜枯萎病菌、苹果轮纹病菌、小麦茎基腐病菌、辣椒疫霉病菌、西瓜炭疽病菌、小麦全蚀病菌和玉米大斑病菌等7种植物病原菌均具有一定的拮抗活性;盆栽试验和田间试验结果表明,杨浦链霉菌WLU210发酵液具有有效防治苦瓜枯萎病和促进苦瓜生长的作用,田间防治效果达68.89%。由此可见,杨浦链霉菌WLU210具有作为防治苦瓜枯萎病生防菌剂研发的潜力。

Abstract: Soil samples were collected from Yellow River wetland at Henan. A dilution coating flat plate method and dual culture assay were applied to isolate actinomycetes strains and evaluate their antimicrobial activity. Finally, one actinomycete strain WLU210 with stable antimicrobial activity were obtained. Based on morphological, physiological and biochemical characteristics along with 16S rRNA gene sequence analysis, the strain WLU210 was identified as Streptomyces yangpuensis. Both S. yangpuensis WLU210 strain and its extract exhibited a broad-spectrum antagonistic activity against seven plant pathogens, such as Fusarium oxysporum f. sp. momordicae, Botryosphaeria dothidea, Fusarium pseudograminearum, Phytophthora capsici, Colletotrichum orbiculare, Gaeumannnomyces graminis var. tritici and Setosphaeria turcica. In addition, the fermentation broth of S. yangpuensis WLU210 could promote the growth of bitter gourd in pot and field tests. Compared to the chemical agent carbendazim with the field control efficacy of 73.89% against bitter gourd Fusarium wilt, strain WLU210 displayed equal control efficacy of 68.89% by root-drenching with 106cfu/mL spore suspension. In summary, S. yangpuensis WLU210 has the potential to be developed into a biocontrol agent for bitter gourd Fusarium wilt control.
