小麦纹枯病拮抗菌HB-10的筛选及其发酵条件优化

Screening of Antagonistic Bacterium HB-10 Against Wheat Sheath Blight and Optimization of Fermentation Conditions

摘要: 为开发对小麦纹枯病有防治效果的生防菌株,本研究筛选出一株对小麦纹枯病病菌有防治效果的HB-10菌株。并对菌株HB-10进行鉴定、发酵优化及盆栽防效试验。结果表明,菌株HB-10对禾谷丝核菌的抑制率为89.72%;通过形态学鉴定和分子鉴定分析得出,菌株HB-10为铜绿假单胞菌Pseudomonas aeruginosa;抑菌谱试验表明,菌株HB-10对烟草赤星病菌、水稻恶苗病菌、稻瘟病菌和油菜菌核病菌都具有较强的拮抗作用。盆栽试验中菌株HB-10对小麦纹枯病相对防效为69.42%。通过正交试验和响应面分析,显示HB-10菌株最适在葡萄糖10 g/L、酵母粉20 g/L、KCl 10 g/L的培养基中生长;最佳的发酵条件为转速215 r/min、温度37℃、接种量为4%、初始pH 7.3、培养时间84 h。经优化后菌株HB-10在24 h的OD600平均值为5.92,比优化前提高了181.8%,对病原菌抑制率也有一定的提升。研究结果为铜绿假单胞菌HB-10生防菌剂的开发及其对小麦纹枯病的防治应用提供了理论依据。

Abstract: To develop biocontrol strains with preventive and therapeutic effects against wheat sheath blight,this study screening HB-10 strain.HB-10 strain exhibited significant control effects against the Rhizoct.onia cerealis.The strain was identified,optimized for fermentation,and evaluated through pot experiments to assess its efficacy.Results showed that HB-10 strain achieved an inhibition rate of 89.72% against Rhizoctonia cerealis.Morphological and molecular identification confirmed HB-10 strain as Pseudomonas aeruginosa.Antimicrobial spectrum analysis demonstrated the strong antagonistic effects of HB-10 strain against various pathogens,including Magnaporthe oryzae,Fusarium moniliforme,Alternaria alternata and Sclerotinia Sclerotiorum.In pot experiments,HB-10 strain exhibited a relative preventive efficacy of 69.42% against wheat sheath blight.Orthogonal tests and response surface analysis revealed the optimal growth medium for HB-10 strain consisted of 10 g/L glucose,20 g/L yeast extract,and 10 g/L KCl.The optimal fermentation conditions included a rotational speed of 215 r/min,temperature of 37℃,inoculum size of 4%,initial pH of 7.3,and a culture time of 84 h.After optimization,the average OD600 value of HB-10 strain at 24 h increased by 181.8%(5.92) compared to the non-optimized condition,resulting in improved inhibition rate against the pathogenic bacteria.These research findings provide a theoretical basis for the development and application of a biocontrol agent using Pseudomonas aeruginosa HB-10,in the prevention and control of wheat sheath blight.
