Identification of Actinomycete Strain QH-16 and Structural Analysis of Its Active Products

Abstract: Antagonistic actinomycetes can produce bioactive compounds with high commercial value and great application potential. In this study, Xanthomonas arboricola pv. pruni was used as the indicator bacteria, and five antagonistic actinomycetes showing inhibition action on pathogen by dual culture assay were screened and strain QH-16 presented the highest antagonistic efficacy. Strain QH-16 was identified by 16S rDNA sequence analysis, morphological observation, cultural and physiological characteristics. The results showed that strain QH-16 was identified as Streptomyces globosus. The filtrate from ferment broth of strain QH-16 could inhibit Fusarium graminearum and Bacillus subtilis, with inhibition bands of 14 mm and 22 mm, respectively. The active ingredients of the fermentation products were extracted by solvent extraction and purified by silica gel column chromatography, thin layer chromatography for preparation (PLC) and preparative analyzed by nuclear magnetic resonanc, the main active product in the fermentation products was identified to be dibutyl phthalate.
